Abstract: The project proposes the developement of interdisciplinary research, both theoretical and experimental, of fundamental aspects of medical applications of positron emitting radioisotops, performed by a consortium of physicists, chemists, biologists and physicians, with the aim of facilitating the implementation of PET and to prepare the conditions of hadron therapy use in treating cancer at national level. One will treat the theoretical and experimental aspects of the production of positron emitting radionuclides (11C, 13N, 15O, 18F, etc.), the ways to incorporate these in radiopharmaceuticals to be used in tomographic imaging based on positron annihilation, as well as the aspects of image reconstruction associated with. The radiolabelling study of specific molecules with biological activity will lead to the selection of some in vivo stable compounds, having high chemical, radiochemical and radionuclidic purity, being able to give informations of functional nature (metabolic processes, blood flow). Image reconstruction algorithms for detection systems of ring and multiple ring types will be presented. The proposed algorithms (Filtered Back Projection and Statistic) will be used to simulate the sinogram acquisition and eventualy the image reconstruction will be obtained. An experimental study aimed at characterising the PET compund interaction with normal and tumoral cells, will be performed. Special techniques from electrophysiology and optical type (TIRF microscopy) will be used to quantitatively measure the state of the system under study and its response to environmental factors (electrical properties of membranes and of the intracellular media, the variation of ions important in cell homeostasis: Ca2+, Na+ and pH). Several effects of accelerated protons and  radiation on cell proliferation will be characterized, with regard to the cellular capacity of repairing DNA strand breaks, the relative density of ionizations produced in DNA by the two different radiations, the kinetics of cell death induced by irradiation, as well as the molecular modifications determined by radiative emission in the process of delayed luminescence.
