Abstract: The development of new therapeutic strategies for anti-cancer care with low toxicity, has become a necessity in the fight against cancer. Recently, there is a greater interest in the use of biomolecules with antitumoral characteristics, such as some antimicrobial peptides (AMPs). Despite their considerable therapeutic potential, there are still some issues related to clinical use of these. An alternative can be found in the use of polyionic transport systems (PIC). Thus, the idea of encapsulating AMPs in these systems can help overcome the obstacles encountered in clinical applications. Also, in order to have a suitable in vitro system, besides cultivating cells in the monolayer, it is necessary to cultivate the cells in a 3D cellular model. The aim of the project is to (i) develop new PIC systems that allow the transport of AMPs to cells, (ii) quantify the antitumor efficiency in 2D and 3D cell systems; (iii) understanding the interactions of the PIC-AMP system with lipid membranes by fluorescence and atomic force studies; and (iv) establishing a composition-activity relationship based on data from cytotoxicity and membrane fluidity studies. For this reason, it will be the first study to consider polyionic materials as delivery systems for AMPs and can have a technological impact by generating new products with increased potential for clinical applications against cancer.
